You will find the product of your box's response and your car's response interacting. That is all. A box that peaks at 40hz in free space might peak at 32hz or 58hz or anything once you put it in a car, especially when you load the sub and/or port. Personally I think the best way is to build your box, put it in the car how you intend to have it for SPL, then block the port. Sweep it and you should have a pretty good idea of your car's peak frequency although remember that the frequency changes as the SPL increases. Quote 10.x volts fo' life!
